The APIs seem to be the source of much that is great and what may ultimately kill Twitter. Minimalism is an under appreciated strength in product design. Twitter not only has an engaged user base, but also a constellation of auxiliary apps hanging off the API. I don't know. I don't think Twitter is going to bleed too much, despite how many bleeding-edge folks are jumping ship for other tools (read: FriendFeed fiends).
